In the tender, ligation of Illumina indexes is requested for each PCR-amplified sample separately. Do you allow alternative method by using barcode-tagged primers and adding indexes to the equimolarly normalized and mixed pool of samples?

As indicated in the Technical Specifications, the use of alternative methods would be acceptable provided that the tenderer can prove that the results would be equivalent to proposed methodology. In principle, we may accept the use of barcode-tagged primers provided that the method does not affect the overall performance of the sequencing, namely the number of obtained reads per sample is still significant to characterize the soil-living communities. The tenderers have to prove this in its offer.
